The Nodame Cantabile anime is coming up this winter season. If it's as good as the drama it'll be a must-see.
The Animation Runner Kuromi OVAs also come to mind, although they lean towards exagerrated slapstick.

This season, there's also a new You're Under Arrest TV anime. Workplace comedy about eccentric traffic cops.
Mahoraba (a series superficially similar to Love Hina) might be worth checking out. It's not wholly realistic but is considerably more down to earth then most comedies of its type. Also has very nice slice-of-life moments.

Well, I've had a look through my collection, and much to my surprise, the only show I've seen that even comes close and has not been mentioned here is Yomigaeru Sora: Rescue Wings. That one is a pretty realistic drama about a JASDF helicopter rescue squad, set at the real JASDF Komatsu base in Ishikawa prefecture.
